Title Establishment and characterization of cell line (OMC-1) derived from mucinous cystadenocarcinoma of human ovary.

Abstract The cell line which was named OMC-1 was established from a well differentiated mucinous cystadenocarcinoma of the ovary in a 75-year-old woman on September 17, 1982. We used a culture medium with D-valine substituted for L-valine in order to inhibit the growth of fibroblast. The OMC-1 cell line has been maintained for 3 years and was subcultivated more than 90 times. The OMC-1 cell line has the following characteristics. 1) The population doubling time was 24 hours. The saturation density was 6.010^6 cells/cm2 and the plating efficiency was 78%. 2) The chromosomal number was widely distributed (43-174), showed aneuploidy and revealed a hypotetraploid tendency. The modal number was 84. 3) The cells were columnar in shape, formed in multilayers, and did not show contact inhibition. The cell had an eccentric nucleus with a few prominent nucleoli. The cytoplasm was granular and finely vacuolated. 4) The cytoplasm contained abundant Golgi apparatus, mitochondria, rough endoplasmic reticulum and secretory granules. The cells had microvilli with core filaments and rootlets at the free cell surface. Junctional complexes were present. 5) In the cytoplasm, Alcian blue, PAS, CEA staining positive substance could be seen. The cells produced CEA and mucinous substance in the culture medium. 6) The cells were transplanted into the subcutis of BALB/c nude mice and produced tumors which were interpreted as well differentiated mucinous cystadenocarcinoma. 7) The morphology of the OMC-1 cell line in vitro and in vivo resembled that of the original tumor. From these findings, the OMC-1 cell line had both morphological and functional characteristics of well differentiated mucinous cystadenocarcinoma of the ovary. Only 3 cell lines have been established from mucinous cystadenocarcinoma of human ovary in the world.
